Restrictions: | The line is available with the following restrictions: 1. This cell line was deposited at the ATCC by Dr. A. Gazdar and Dr. J. Minna and is provided for research purposes only. Neither the cell line nor products derived from it may be sold or used for commercial purposes. Nor can the cells be distributed to third parties for purposes of sale, or producing for sale, cells or their products. The cells are provided as service to the research community. They are provided without warranty of merchantability or fitness for a particular purpose or any other warranty, expressed or implied. 2. Cytogenetic Analysis: | modal number = 61; range = 55 to 64. This is a hypotriploid human cell line. The modal chromosome number is 61, occurring in 28% of cells. The rate of higher ploidies is 1.2%. Twelve marker chromosomes are common in these cells. These include t(7q,16q), t(5p,7p), t(4q,9q) and 9 others. A large number of DM's is found in every cell. The N21 is absent; and N9, N11, N13, N20, N22 and X are single-copied. No Y chromosome was detected in the Q-banded slide preparation. |

Comments: | This line was derived from cells present in ascites fluid obtained from a patient after treatment with 5-fluorouracil. The cells contain Dopa decarboxylase and, unlike other colorectal lines, contain cytoplasmic dense core granules characteristic of endocrine secretion. The cells do not express the TAG-72 or CA19-9 antigens nor do they produce carcinoembryonic antigen (CEA). |
Propagation: | ATCC complete growth medium: The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%. Atmosphere: air, 95%; carbon dioxide (CO2), 5% Temperature: 37.0°C |
Subculturing: | Protocol: Cultures can be maintained by the addition of fresh medium or replacement of medium Cultures can be established by centrifugation with subsequent resuspension at 4 to 8 X 10(5) viable cells/ml. Subc*tion Ratio: A subc*tion ratio of 1:3 to 1:6 is recommended Medium Renewal: Every 2 to 3 days |
Preservation: | Freeze medium: Complete growth medium supplemented with 5% (v/v) DMSO Storage temperature: liquid nitrogen vapor phase |
Related Products: | Recommended medium (without the additional supplements or serum described under ATCC Medium):ATCC 30-2001 recommended serum:ATCC 30-2020 |
