淀粉|酶是一種水解酶，它能夠作用于淀粉分子中的a-1，4-糖苷鍵，將其分解為葡萄糖。a-淀粉|酶可以作用于直鏈淀粉和支鏈淀粉，切開淀粉鏈?zhǔn)蛊滢D(zhuǎn)化成麥芽糖、麥芽三糖、葡萄糖和極限糊精。對(duì)哺乳動(dòng)物來說，a-淀粉|酶是主要的消化酶，其含量增高可能與涎腺損傷、腮腺炎、胰腺炎或腎衰竭等疾病有關(guān)。在科研和藥物開發(fā)中，簡單、自動(dòng)化的a-淀粉\酶檢測(cè)手段是zui為理想的。本公司的的a-淀粉|酶檢測(cè)包含兩個(gè)步驟：(1) a-淀粉|酶將樣品中的淀粉水解，其產(chǎn)物在a-葡糖苷酶和葡糖氧化酶的作用下，轉(zhuǎn)變?yōu)槠咸烟呛碗p氧水。(2) 利用一種比色試劑，檢測(cè)出雙氧水濃度。
測(cè)定方法 比色法（585nm）定量測(cè)定α-淀粉|酶活性。
產(chǎn)品特點(diǎn)
檢測(cè)敏感、準(zhǔn)確：用96孔板檢測(cè)，a-淀粉|酶活性檢測(cè)范圍為0.3 - 50 U/L。
操作簡便：整個(gè)檢測(cè)過程為：加入一種工作試劑并反應(yīng)15分鐘，然后加入檢測(cè)試劑再反應(yīng)20分鐘后，在585nm下讀取吸光度。
產(chǎn)品用途 直接檢測(cè)血液、唾液、尿液、谷物和其他農(nóng)產(chǎn)品中a-淀粉|酶的活性。
儲(chǔ)藏條件： 冰運(yùn)。底物和檢測(cè)試劑在4°C的環(huán)境下保存，其它試劑在-20°C下保存。
Description Amylase belongs to the family of glycoside hydrolase enzymes that break down starch into glucose molecules by acting on α-1,4-glycosidic bonds. The α-amylases (EC 3.2.1.1) cleave at random locations on the starch chain, ultimay yielding maltotriose and maltose, glucose and "limit dextrin" from amylose and amylopectin. In mammals, α-amylase is a major digestive enzyme. Increased enzyme levels in humans are associated with salivary trauma, mumps due to inflammation of the salivary glands, pancreatitis and renal failure.Simple, direct and automation-ready procedures for measuring amylase activity are very desirable. our EnzyChromTM a-amylase assay method involves two steps: (1). α-amylase in the sample hydrolyzes starch and the product is rapidly converted to glucose by α-glucosidase and hydrogen peroxide by glucose oxidase; (2). hydrogen peroxidconcentration is determined with a colorimetric reagent.
Applications Direct assays of α-amylase activity in blood, saliva, urine, grains and other agricultural samples.
Key features Sensitive and accurate. Linear detection range 0.3 to 50 U/L α-amylase in 96-well plate assay.
Convenient. The procedure involves adding a single working reagent, incubation for 15 min, followed by the detection reagent and a 20-min incubation and reading the optical density at 585 nm.
Storage conditions. Kit is shipped on dry ice. Store Substrate and Detection Reagent at 4°C and others at -20°C. Shelf life of at least 6 months (see expiry dates on labels).
