近期，中國(guó)醫(yī)學(xué)科學(xué)院皮膚病研究所姚煦教授團(tuán)隊(duì)、復(fù)旦大學(xué)附屬華山醫(yī)院李巍教授團(tuán)隊(duì)及香港城市大學(xué)董立新教授團(tuán)隊(duì)合作在 Cell Host & Microbe（IF=20.6）上發(fā)表題為“Multi-omics Signatures Reveal Genomic and Functional Heterogeneity of Cutibacterium acnes in Normal and Diseased Skin."（多組學(xué)特征揭示正常和患病皮膚中痤瘡丙酸桿菌的基因組和功能異質(zhì)性）的研究，在該項(xiàng)目中，研究團(tuán)隊(duì)從1234個(gè)分離基因組中證明了C. acnes的個(gè)體和生態(tài)位異質(zhì)性。皮膚疾?。裾詈宛畀彛┖蜕眢w部位決定了C. acnes的基因組差異，這種差異源于水平基因轉(zhuǎn)移和選擇壓力。與表皮葡萄球菌（Staphylococcus epidermidis）相比，C. acnes具有獨(dú)-特的代謝功能，抗生素耐藥基因和毒力因子較少，基因組更穩(wěn)定。綜合基因組、轉(zhuǎn)錄組和代謝組分析在菌株水平揭示了C. acnes的功能特征。與轉(zhuǎn)錄組特征一致，C. acnes在富含皮脂的環(huán)境中對(duì)角質(zhì)形成細(xì)胞產(chǎn)生毒性和促炎作用。L-肌肽（L-carnosine）是一種抗氧化應(yīng)激代謝物，在濕疹的代謝組中上調(diào)并減輕皮膚炎癥?？偟膩?lái)說(shuō)，該研究揭示了基因和微環(huán)境對(duì)C. acnes功能的共同影響。利用從健康皮膚、濕疹（AD）和痤瘡患者皮膚分離的痤瘡丙酸桿菌（Cutibacterium acnes, C. acnes）菌株進(jìn)行了全基因組測(cè)序、轉(zhuǎn)錄組測(cè)序、代謝組測(cè)序以及功能研究，研究團(tuán)隊(duì)首-次從單菌株層面揭示了C. acnes的基因和功能異質(zhì)性。通過(guò)深入地了解宿主和皮膚微生物群之間的復(fù)雜關(guān)系，有望能找到增進(jìn)皮膚健康和改善皮膚病管理的靶點(diǎn)，也為將來(lái)探索C. acnes的臨床應(yīng)用奠定了基礎(chǔ)。
