Anti asialo GM1 (Rabbit) NK細(xì)胞抑制劑
產(chǎn)品介紹:
Anti asialo GM1的抗血清可以與NK細(xì)胞(自然殺傷細(xì)胞)結(jié)合,抑制NK細(xì)胞非特異性殺傷靶細(xì)胞的活性。靶細(xì)胞包括腫瘤細(xì)胞,同種異體移植的器官、組織等??捎糜谒幬锖Y選、器官抑制、自身免疫疾病研究等。NK細(xì)胞屬于天然免疫細(xì)胞,具有免疫監(jiān)視功能。
特異性:小鼠、大鼠的NK細(xì)胞,小鼠單核細(xì)胞(不含有NK細(xì)胞的肝臟細(xì)胞,骨髓,胎兒肝臟細(xì)胞,裸鼠巨噬細(xì)胞的脾細(xì)胞),胎兒T淋巴細(xì)胞(12天)。
◆研究糖尿病的免疫治療:
據(jù)報(bào)道,抗細(xì)胞凋亡細(xì)胞因子IL-15具有促進(jìn)NK細(xì)胞和T細(xì)胞(包括調(diào)節(jié)T細(xì)胞,Tregs)存活和激活上述兩種細(xì)胞活性的功能。在患有糖尿病的NOD小鼠體內(nèi)反復(fù)注射IL-15。如果每天一次,注射2周,既不*也不加速未處理NOD小鼠患糖尿病的情況。但是NK細(xì)胞活性 喪失的NOD小鼠折射IL-15可以明顯減少糖尿病的發(fā)病率和延遲糖尿病發(fā)病時(shí)間,但是NK細(xì)胞活性喪失不注射IL-15的小鼠依然病發(fā)糖尿病。在用IL-15處理,NK細(xì)胞活性喪失的NOD小鼠體內(nèi)產(chǎn)生的保護(hù)效應(yīng)與CD4+CD25+Tregs免疫抑制活性增加有關(guān)。在該項(xiàng)實(shí)驗(yàn)中,用Anti AsGM1注射小鼠,抑制NK細(xì)胞活性,導(dǎo)致NOD小鼠NK細(xì)胞活性缺失。
◆研究敗血癥中NK細(xì)胞的功能:
NK細(xì)胞和NKT細(xì)胞都屬于天然免疫系統(tǒng),但在細(xì)菌引起的敗血癥中這兩種細(xì)胞的作用是有爭(zhēng)議。用Anti Asialo GM1處理小鼠,成為NK細(xì)胞活性抑制的小鼠;用CD1d單抗處理小鼠,成為NKT細(xì)胞活性抑制 的小鼠;同時(shí)用同型對(duì)照單抗處理小鼠,作為陰性對(duì)照。之后這些小鼠感染肺炎鏈球菌。分析血清和組織樣品中細(xì)菌的含量,細(xì)胞因子,脾細(xì)胞凋亡率、用流式鑒定細(xì)胞,同時(shí)也分析脾細(xì)胞miRNA的表達(dá)。結(jié)果發(fā)現(xiàn)NK細(xì)胞活性抑制的小鼠延長(zhǎng)了存活率。一單NKT細(xì)胞激活抑制,與其他細(xì)胞群相比,脾臟的NK(CD3-/NK1.1+)細(xì)胞數(shù)目增加。NKT細(xì)胞活性抑制導(dǎo)致小鼠體內(nèi)的細(xì)菌含量升高,導(dǎo)致血清和脾細(xì)胞FN-γ水平升高??笴D1d處理小鼠的脾細(xì)胞miRNA分析顯示miR-200C和miR-29a下調(diào),miR125a-5p上調(diào)。這些變化發(fā)生在NK細(xì)胞活性抑制后,NK細(xì)胞刻個(gè)與肺炎鏈球菌肺炎的死亡率有關(guān)。抑制NKT細(xì)胞的激活導(dǎo)致(CD3-/NK1.1+)細(xì)胞數(shù)目增加,F(xiàn)N-γ水平升高,改變脾細(xì)胞miRNA的表達(dá)。
◆研究自身免疫性疾病的NK細(xì)胞功能
NK細(xì)胞是先天免疫淋巴細(xì)胞,在先天免疫和適應(yīng)性免疫中均起著關(guān)鍵性作用。它在小鼠體內(nèi)通過(guò)NK細(xì)胞消耗 Abs,特別是抗唾液酸GM1 (ASGM1),起關(guān)鍵作用。此次研究中,我們通過(guò)整個(gè)嗜堿性粒細(xì)胞群體組成如表達(dá)NK細(xì)胞一樣,來(lái)表達(dá)ASGM1以及CD49b (DX5) ,并通過(guò)抗ASGM1在體內(nèi)通過(guò)嗜堿性粒細(xì)胞消耗的抗FceRlaAb有效地消融??笰SGM1介導(dǎo)的嗜堿性粒細(xì)胞的消耗與各種小鼠品系的NK細(xì)胞消耗有關(guān),與K1同種異型和MHC H2單體型(包括C57BL/6, BALB/c, C3H和小鼠A/J)無(wú)關(guān)。這些結(jié)果將嗜堿性粒細(xì)胞定義為以前未被認(rèn)識(shí)到的抗ASGM1介導(dǎo)細(xì)胞消耗的靶標(biāo),并引起人們對(duì)嗜堿性粒細(xì)胞的關(guān)注,而不是NK細(xì)胞或除了NK細(xì)胞外對(duì)抗ASGM1處理的小鼠中觀察到的某些表型
◆研究腫瘤的免疫治療
腫瘤的發(fā)展過(guò)程包括血管新生,腫瘤尺寸增大和腫瘤轉(zhuǎn)移。使用CXCL14/BRAK (CXCL14)轉(zhuǎn)基因小鼠研究過(guò)度表達(dá)的CXC趨化因子配體14 (CXCL14)在上述生物學(xué)現(xiàn)象中的作用。與野生型小鼠相比,轉(zhuǎn)基因小鼠經(jīng)AOM/DSS誘導(dǎo)的結(jié)腸血管新生的發(fā)病率顯著低很多。腫瘤細(xì)胞注射到這些小鼠后,腫瘤尺寸發(fā)生變化。與野生型的相比,轉(zhuǎn)基因小鼠肺中轉(zhuǎn)移小瘤的數(shù)目明顯少,腫瘤的尺寸也明顯變小。在注射腫瘤前后注射Anti Asialo GM1減少了CXCL14在腫瘤生長(zhǎng)方面和腫瘤轉(zhuǎn)移的抑制效應(yīng),表明激活的NK細(xì)胞在CXCL14介導(dǎo)的抑制腫瘤生長(zhǎng)和轉(zhuǎn)移期間起重要作用。CXCL14也表達(dá)在 B16黑色素瘤細(xì)胞內(nèi)表明NK細(xì)胞在腫瘤轉(zhuǎn)移方面的重要性。而且轉(zhuǎn)基因小鼠在注射腫瘤細(xì)胞后的存活率顯著增長(zhǎng)。這些轉(zhuǎn)基因小鼠沒(méi)有明顯的不正常,預(yù)計(jì)腫瘤抑制/預(yù)防方面CXCL14起著重要作用
